D'origine italo-américaine, il se destine tout d'abord au séminaire. Il entre en 1963 à l'Université de New York et y réalise deux courts métrages. Tour à tour réalisateur publicitaire, assistant ou monteur, il travaille avec Michael Wadleigh (Woodstock) et François Reichenbach. En 1973, il se fait remarquer avec Mean Streets, description d'une jeunesse new-yorkaise qu'il connaît bien. Toute la ligne moralisatrice de Scorsese est dans ce film. Avec la complicité de son scénariste Paul Schrader, il décrit un univers violent et corrompu où les hommes se démènent, cherchant sans cesse la rédemption. Alice n'est plus ici est un aparté dans son monde viril décrivant une mère qui quitte la maison pour tenter une carrière artistique. Les meilleures réussites de Scorsese se concrétisent avec Robert De Niro, son double et son acteur fétiche. Qu'il soit chauffeur de taxi témoin de la décadence New-yorkaise dans Taxi Driver (Palme d'or à Cannes 1976), saxophoniste dans New York, New York, Jack La Motta dans Raging Bull, le groupie de Jerry Lewis dans La Valse des Pantins, mafioso dans Les Affranchis et Casino ou bagnard sanguinaire dans Les Nerfs à Vif (remake du film de J. Lee Thompson), Robert De Niro entraîne le spectateur vers la grandeur et la déchéance des paumés et des maginaux entre affliction et rédemption. Le cinéaste enchante avec une commande comme After Hours à l'univers kafkaïen. Il fait scandale en 1987 avec La Dernière Tentation du Christ alors qu'il ne fait que traduire les préoccupations présentes dans le roman de Katsantakis. Sa quête mystique l'entraîne vers la nostalgie du Temps de l'Innocence et la méditation orientale dans Kundun. Il revient à ses racines avec À tombeau ouvert sorte de Taxi Driver, 20 ans après débouchant sur le constat pessimiste. Pourtant Scorsese est très attaché à ses racines témoignant avec L'Album de Martin Scorsese, ironisant avec New York Stories, retraçant l'histoire de sa ville dans Gangs of New York. Adulé dans le monde entier (penser à Benigni se jettant à ses pieds à Cannes), Scorsese fait partie des plus grands créateurs du cinéma américain.
