Débutant comme mannequin à seize ans, elle fait ses débuts à la Vitagraph avant de rejoindre la Biograph de Griffith qui lui offre son premier grand rôle dans Son éveil (Her Awakening) en 1911. Elle fait la connaissance de Mack Sennett avec qui elle fonde les Keystone Studios et impose Charles Chaplin, malgré les réticences de Sennett. Le couple burlesque s'impose dans une vingtaine de films dans lesquels elle vole souvent la vedette à Charlot (Gentlemen of Nerve, His trysting Place). Elle est une des premières actrices à prendre des tartes à la crême en plein visage et effectuer des cabrioles toutes dentelles dehors. Comme ses homologues masculins, Harold Lloyd et Buster Keaton, Mabel est la véritable auteur de ses films qu'elle écrit, produit et réalise en sous-main surtout après sa séparation avec Mack Sennett et la signature de son contrat en 1918 avec Samuel Goldwyn. Elle tourne plusieurs films avec Roscoe Arbuckle, le burlesque obèse connu sous le nom de Fatty (Mabel and Fatty's Walsh Day, Mabel and Fatty's simple Live). Elle co-signe plusieurs films avec F. Richard Jones (Molly O', Suzanna, The Extra Girl) mais se brouille rapidement. Sa vie dissolue est émaillée de scandales comme l'assassinat du réalisateur et producteur William Desmond Taylor dont elle partage la vie et le meurtre de Joe Kelly abattu par son chauffeur. Dans les années vingt, elle rejoint les studios d'Hal Roach. Elle dirige plusieurs films co-écrits par Stan Laurel et réalisés par Leo McCarey (Should Men walk home?). En 1926, elle épouse l'acteur Lew Cody mais sa santé décline. Atteinte de tuberculose, elle meurt à Monrovia à l'âge de 37 ans. Superstar du muet, elle sert de modèle à Billy Wilder pour le personnage de Norma Desmond (composé de Normand et W. Desmond Taylor) dans Boulevard du Crépuscule.
Mabel Normand est décédée à Monrovia, le 23 février 1930.
