Issu de milieux aisés, fils d'un professeur de Harvard, il débute comme journaliste et s'intéresse au théâtre. Il réalise des films courts d'inspiration surréaliste comme Soul of the Cypress, inspiré du mythe d'Orphée avec sa première épouse Chase Harringdine ou Dans Macabre. Il vient en France où il collabore au Ballet mécanique de Fernand Léger en 1924 où il apparaît auprès de sa deuxième Kathrin Hawley Murphy. S'il est le véritable moteur de l'œuvre, le peintre en tirera toute la gloire. Revenu aux États-Unis, il signe des films musicaux interprétés par des noirs célèbres comme Bessie Smith dans Saint Louis Blues ou Paul Robeson dans The Emperor Jones. En 1932, il collabore avec l'artiste mexicain David Alfaro Siqueiros. L'un de ses derniers films importants, Main Street Lawyer retrace un épisode de la vie d'Abraham Lincoln, incarné par Edward Ellis. Cet esthète qui fut un des premiers cinéastes indépendants d'Hollywood, s'installe de 1940 aux années soixante au Mexique en compagnie de sa nouvelle femme Virginia et devient le directeur d'un complexe hôtelier en Californie. Longtemps ignoré, il est redécouvert grâce à la biographie Dudley Murphy, Hollywood Wild Card de Susan Delson qui met en lumière les différents aspects novateurs du réalisateur.
Dudley Murphy est décédé à Mexico, le 22 février 1968.
