Il découvre le cinéma comme assistant de Pupi Avati et Marco Risi. Il étudie l'écriture de scénario au Centre Expérimental de Cinématographie de Rome et travaille pour la RAI. Il signe un sketch du film collectif Intolérance et plusieurs épisodes du feuilleton Une Place au Soleil (Un Posto al Sole). Avec Ecco Fatto, il dessine les thèmes dominants de ses films, l'amour, la jalousie, le rôle du père et de la mère en décrivant la relation entre un jeune étudiant immature et une femme plus âgée et indépendante. Comme toi s'intéresse aux flirts d'adolescents, le héros en est Silvio Muccino, le propre frère du réalisateur. Juste un Baiser allie le marivaudage et la comédie romantique. Véritable triomphe en Italie, il fait l'objet d'un remake à Hollywood par Tony Goldwyn. Cette percée lance la carrière internationale de Muccino qui confirme ses belles intentions avec le touchant À la Recherche du Bonheur qui met en présence un surprenant Will Smith en compagnie de son fils Jaden. Un père plaqué par sa femme se retrouve à rechercher désespérement un job et finit dans une semi-clochardise avant de s'en sortir grâce à la confiance de son gamin. Sentimentalisme et optimisme font de Muccino le plus américain des jeunes cinéastes transalpins.
