Il grandit dans les milieux du spectacle, ses parents étant auteurs dramatiques. Fondateur en 1912 de la société Lasky Feature Play Company avec Jesse L. Lasky et Samuel Goldfish (alias Goldwyn), il connaît un très vif succès dès sa première réalisation The Squaw Man en 1913. Spécialisé dans le western (La Rose du ranch, La Captive), l'aventure exotique (L'Arabe, Forfaiture) et la comédie de mœurs sexy (L'échange, L'admirable Crichton, Le Cœur nous trompe, tous avec Gloria Swanson), il trouve définitivement sa voie dans la reconstitution historique (Jeanne d'Arc) ou biblique (Les dix Commandements). Selon sa formule " Du sexe, du sang et de la bible " (sex, blood and Bible), il réalise des superproductions à la mise en scène gigantesque, aux figurants innombrables traitant de sujets historiques saupoudrées d'un érotisme sous-jacent. Ses œuvres parlantes sont significatives de son style : Le Signe de la Croix, Cléopatre (Claudette Colbert dans son bain de lait), Samson et Dalila (avec la sculpturale Hedi Lamarr) et sa deuxième version des Dix Commandements avec Charlton Heston. Mais c'est avec Gary Cooper qu'il donnera le meilleur de lui-même avec des westerns (Une aventure de Buffalo Bill, Pacific Express, Les Tuniques écarlates, Les Conquérants du Nouveau Monde) ou son épopée médicale (L'Odyssée du Docteur Wassel). Billy Wilder lui rend hommage dans Boulevard du Crépuscule où Cecil B. De Mille dirige une dernière fois Gloria Swanson. Son dernier projet, Les Boucaniers, remake de son film de 1937 avec Fredric March sera finalement tourné par son gendre, Anthony Quinn. Cecil B. De Mille avec une œuvre inégale mais d'une grande force visuelle reste un des plus célèbres cinéastes du cinéma américain.
Cecil B. De Mille est décédé à Hollywood, le 21 janvier 1959.
