Brillant universitaire, il est remarqué par Spielberg qui lui confie les scénarios de ses productions : Gremlins de Dante, Les Goonies de Donner, Le Secret de la pyramide (Young Sherlock Holmes) de Levinson. Il participe au scénario d'Indiana Jones et la dernière croisade. Il signe sa première réalisation en 1987 avec Nuit de Folie, une comédie typiquement américaine mais brillamment enlevée. Heartbreak Hotel s'inscrie dans les films ados façon John Hughes. Avec son troisième film, Maman j'ai raté l'avion dont il tirera une suite un an plus tard, il décroche le jackpot. Scénario bien ficelé, personnage ultra stéréotypé, un gamin au jeu épouvantable mais qui devient une mégastar (Culkin), ce divertissement lance Columbus sur les rails du succès. Madame Doubtfire, comédie propre à mettre en valeur les talents de Robin Williams confirme l'aura de ce golden boy. Si sa version américaine de Neuf mois de Braoudé n'apporte que le plaisir de voir de grands acteurs, Ma meilleure ennemie prouve qu'il peut aussi faire mouche dans le mélodrame. Enfin Harry Potter devient la synthèse de son cinéma : gros moyens, scénario béton, acteurs solides, divertissement assuré bien que ne reculant devant aucun effet facile.
