Il débute dans les années 70 dans différentes séries avant de collaborer avec John Carpenter pour Halloween, où il interprète un petit rôle. Les deux complices scénaristes se retrouvent pour Christine d'après Stephen King et New York 1997. Il signe son premier film personnel en 1984 avec The Last Starfighter, un film d'anticipation plutôt réussi. La diversité de ses films est importante passant du film de danse (Tap Dance) à la comédie pour enfants (Denis la Malice, Major Payne), la fantaisie fantastique (The Boy who could fly) et le polar (Mr Wrong). Éclectique, Nick Castle est très apprécié aux Etats-Unis mais demeure plus confidentiel en Europe.
