D'origine écossaise, né au Canada, il arrive aux États-Unis en 1896. Il tente sa chance à Hollywood et sous la houlette de Cecil B. De Mille et surtout de Thomas H. Ince, il dirige des secondes équipes pour de vigoureux westerns. Il se voit confier par Ince la réalisation d'une série "japonaise" avec Sessue Hayakawa (Honneur japonais, The Ambassador's Envoy). Il signe ensuite totalement ou en partie les meilleurs films de William S. Hart (The Bargain, Le Disciple, Le Justicier). Il amène le western à son apogée en réalisant les scènes les plus spectaculaires de Civilisation que co-signe Ince. Ses contemporains sont en admiration pour Le Marché aux Chimères et Carmen du Klondyke qu'interprète Clara Williams. Dans les années 20, il signe également de grands mélodrames comme La Flamme du Désert avec Geraldine Farrar, La Lutte éternelle avec Renée Adorée, La Rançon avec Wallace Beery, Broken Barries avec Norma Shearer et Body and Soul avec Lionel Barrymore. Il ne signe malheureusement aucune œuvre majeure à l'avènement du parlant (à l'exception de The Moonstone) et disparaît prématurément. Reginald Barker fait partie de ces grands cinéastes du muet injustement oubliés dont l'œuvre est redécouverte récemment.
Reginald Barker est décédé à Los Angeles, le 23 février 1945.
