Acteur dès l'âge de 8 ans, il effectue des tournées avec des spectacles chantants et dansants. Il est ensuite embauché à la Selig, avant de passer chez Mack Sennett, puis à la Triangle. Il crée un personnage comique obèse, très à l'aise cependant dans des scènes de poursuite et très agile malgré cette corpulence qui lui vaudra le surnom de "Fatty". Avec l'apport de scénarios de Al Saint-John, son neveu, d'Alice Lake et surtout de Buster Keaton, il dirige lui-même ses films à partir de 1917. Une rivalité avec son neveu le renvoie au rôle de simple acteur dans des longs métrages signés Taurog ou Henaberry.Le scandale provoqué par la mort de l'actrice Virginia Rappe au cours d'une party organisée par Arbuckle lui coûtera sa carrière et vaudra l'emprisonnement. Engagé par Keaton, il signera encore quelques scénarios sous des noms d'emprunt (Willy B. Good, William Goodrich). Son nom reste attaché à l'âge d'or du burlesque américain.
Fatty Arbuckle est décédé à Los Angeles, le 29 juin 1933.
